益甘舒抗乙醇诱导斑马鱼兴奋期醉酒的作用研究Effect of Yiganshu on the Anti-alcohol in Zebrafish

摘要(Abstract):
目的评价药食同源健康产品益甘舒对斑马鱼的解酒作用,为益甘舒的应用提供理论依据。方法随机选取240尾受精后5 d(5 dpf)野生型AB品系斑马鱼于六孔板中,每孔(实验组)均处理30尾斑马鱼。设立益甘舒不同浓度组、阳性对照药"RU21"组、正常对照组和模型对照组。益甘舒与阳性对照药"RU21"分别处理24 h后,每个实验组随机选取10尾斑马鱼于96孔板中,每孔1尾斑马鱼,容量为200μL,除正常对照组外,其余实验组均给予1.5%乙醇诱导建立斑马鱼兴奋期醉酒模型,用运动/行为分析记录仪拍摄并记录斑马鱼在1 h内的总运动距离。通过对总运动距离的统计分析,评价益肝舒对斑马鱼酒精中毒模型的影响。结果模型对照组斑马鱼的总运动距离与正常对照组比较P<0.001,表明模型建立成功。与模型对照组比较,阳性对照药"RU21"组、益甘舒500、1 000和2 000μg·m L-1浓度组对乙醇诱导的兴奋期醉酒模型斑马鱼均有显著的醒酒作用(P<0.05),且剂量越大作用越明显。结论在本实验浓度条件下,益甘舒对乙醇诱导的兴奋期醉酒模型斑马鱼具有明显的解酒作用。
OBJECTIVE To evaluate the anti alcohol effect of Yiganshu on zebrafish and provide theoretical basis for its application.METHODS A total of 240 wild type AB strains of zebrafish( 5 dpf) were randomly selected on the six hole plate,and 30 zebrafish( experimental group) were treated with each hole. Different concentrations of Yiganshu group,positive control drug "RU21" group,normal control group and model control group were established. After 24 hours of treatment with Yiganshu and positive control drug " RU21",10 zebrafish in each experimental group were randomly selected to feed in 96 well plate,one per hole,with a capacity of 200 μL.In addition to the normal control group,the other experimental groups were given 1.5% ethanol to induce the establishment of zebrafish intoxication model.The total movement distance of zebrafish in one hour was recorded by using motion/behavior analysis recorder. The influence of Yiganshu on the model of zebrafish alcoholism was evaluated by statistical analysis of the total exercise distance. RESULTS The total moving distance of zebrafish in the model control group was significantly lower than that in the normal control group( P < 0. 001).Compared with the model control group,the positive control group,Yiganshu middle and high concentration group had significant sobering effect on the ethanol poisoned zebrafish model( P<0.05),with dose response association. CONCLUSION Yiganshu has an obvious effect on the alcohol-induced intoxication model zebrafish.
