摘要(Abstract):
目的建立快速液相色谱-串联质谱法(RRLC-MS/MS)同时测定罂粟壳药材中5种生物碱(可待因、吗啡、罂粟碱、蒂巴因和那可丁)的含量。方法色谱柱为菲罗门Luna C18(150 mm×2.00 mm,3μm),甲醇-10 mmol·L~(-1)乙酸氨水为流动相梯度洗脱,电喷雾离子源正离子模式,选择性反应监测。结果 5种目标成分在各自线性范围内线性关系良好(r>0.997 6),加样回收率在96.1%~99.4%之间,RSD为2.5%~5.0%。5种生物碱总量为0.505%~0.760%,依次为吗啡>那可丁>可待因>罂粟碱>蒂巴因。结论本方法能快速、简便、有效地测定罂粟壳药材中多种指标性成分,为临床应用的质量标准提供参考。
OBJECTIVE To develop a method for the determination of five alkaloids( codein,morphine,papaverine,thebaine and narcotine) in Pericarpium Papaveris using rapid resolution liquid chromatography coupled to triple quadrupole mass spectrometry( RRLC-MS/MS). METHODS The separation was performed on a Phenomenex Luna C18 column( 150 mm×2.0 mm,3 μm),using methanol -10 mmol·L~(-1) ammonium acetate as mobile phase with gradient elution. The five target components were detected in multiplereaction monitoring( MRM) mode by mass spectrometry with electrospray ionization( ESI) source operated in positive ionization mode.RESULTS Good linearity was achieved in their respective linear ranges( r>0.997 6),and the recoveries ranged from 96.1%-98.4%with the RSDs ranged from 2.5%-5.0%. The quantitative results showed that the overall content of five alkaloids was 0.505%-0.760%.The largest content was morphine,with the second narcotine,and smallest thebaine. Codein and papaverine had different size.CONCLUSION The method is rapid,simple and effective for the target components quantification in Pericarpium Papaveris. It is supplied for quality standard in clinic study.
