基于均匀设计-偏最小二乘回归优化四黄止痛喷雾剂中黄连与黄柏提取工艺的研究

摘要(Abstract):

目的优选四黄止痛喷雾剂中黄连、黄柏的提取工艺。方法实验采用均匀设计法和偏最小二乘回归分析法,以乙醇浓度、乙醇用量、提取时间、提取次数为主要考察因素,以盐酸小檗碱的转移率为指标优化提取工艺。结果优选工艺条件为:药材饮片加入4倍量56%乙醇,煎煮3次,每次105 min,在此条件下,盐酸小檗碱转移率的平均值为Y=88.93%。结论所优选的工艺可行、重复性好。
OBJECTIVE To optimize extraction process of Rhizome Coptidis and Cortex Phellodendri in Sihuang Zhitong spray.METHODS The experiment combined uniform design and Partial Least Squares(PLS).The ethanol concentration,solvent dosage,extracting time,and extracting times were used as main test factors.The transfer rate of Berberine hydrochloride was used as investigation index to optimize the extraction process.RESULTS The optimal conditions were as follows: adding 4 times the amount of herbal medicine 56% ethanol extract,boiling 3 times and each time for 105 min.Under these conditions,the average yield of total Berberine hydro chloride was 88.93%.CONCLUSION The optimized extraction process is feasible with good repeatability.
