摘要(Abstract):
目的研究鸡骨草的不同提取方法对非酒精性脂肪肝(NAFLD)的改善作用。方法长期饲喂高脂饲料建立大鼠NAFLD模型,给予鸡骨草水提取物及醇提取物,测定大鼠肝组织甘油三酯(TG)、胆固醇(TC)、游离脂肪酸(FFA)和血清炎症细胞因子肿瘤坏死因子-α(TNF-α)、白介素-1(IL-1)水平。结果鸡骨草水提物及鸡骨草50%醇提物高剂量组显著降低TG、TC及FFA水平(P<0.01),鸡骨草50%醇提物低剂量组降低TC及FFA水平(P<0.05或P<0.01);鸡骨草水提物及50%醇提物高剂量组显著降低IL-1、TNF-α水平(P<0.01),鸡骨草水提物低剂量组降低IL-1、TNF-α水平(P<0.05)。结论鸡骨草水提物改善脂质代谢紊乱及调控炎症细胞因子作用较鸡骨草50%醇提物显著,而以鸡骨草水提物高剂量组改善效果最好,本研究为阐明含鸡骨草制剂防治肝损伤相关疾病的药效物质基础提供理论与实验依据。
OBJECTIVE To study the effects of Abrus mollis Hance by different extraction methods on non-alcoholic fatty liver disease( NAFLD). METHODS The NAFLD rat model was established by long-term feeding of high-fat diets. The level of TG,TC and FFA in liver tissues,and TNF-α 、IL-1 in serum lipids was determined after taken orally aqueous and alcohol extract of Abrus mollis Hance. RESULTS Aqueous extract group and 50% alcohol high-dose extract group significantly reduced the level of TG,TC and FFA( P<0.01),and 50% alcohol low-dose extract group reduced the level of TC and FFA( P<0.05,P<0.01); Aqueous extract group and 50% alcohol high-dose extract group significantly reduced the level of TNF-α and IL-1( P < 0. 01),and aqueous low-dose extract group reduced the level of TNF-α and IL-1( P<0.05). CONCLUSION The effects of aqueous extract of Abrus mollis Hance on lipid metabolism disorders and regulation of inflammatory cytokines are more significant than 50% alcohol high-dose extract groups,and the aqueous high-dose extract group shows the best effect. This research provides a theory and experiment basis for Abrus mollis Hance composition on prevention and cure hepatic injures disease.
