HPLC法测定大豆肽蛋白质粉中β-羟基-β-甲基丁酸的含量

摘要(Abstract):

目的建立测定大豆肽蛋白质粉中β-羟基-β-甲基丁酸含量的高效液相色谱(HPLC)法。方法色谱柱为Phenomenex Uranus C_(18)柱(250 mm×4.6 mm,5μm),流动相为0.01 mol·L~(-1)庚烷磺酸钠溶液-乙腈(95:5,V/V,10%磷酸溶液调节pH值为3.0),柱温为35℃,流速为0.5 mL·min~(-1),样品进样量为5μL,检测波长为214 nm。结果本方法中β-羟基-β-甲基丁酸的线性范围为0.088 67~0.886 7 mg·mL~(-1),回归方程:Y=3.566X-0.006(r~2=1.000);方法的加样回收率为95.19%~103.70%。结论该方法操作简便、结果准确,适用于测定大豆肽蛋白质粉中β-羟基-β-甲基丁酸的含量。
OBJECTIVE To establish a method for quantitative determination of β-hydroxy-β-methylbutyrate in soy peptide protein complex powder by high performance liquid chromatography. METHODS Phenomenex Uranus C_(18)column( 250 mm×4.6 mm,5 μm) was used,with 0.01 mol·L~(-1)sodium heptanesulfonate solution-acetonitrile( 95 :5,V/V10% phosphoric acid solution adjusted to pH 3.0) as the mobile phase. The column temperature was 35 ℃,the flow rate was 0. 5 mL·min~(-1),sample injection volume was 5 μL,the detection wavelength was 214 nm. RESULTS The linear range of β-hydroxy-β-methylbutyrate was 0. 088 67-0. 886 7 mg·mL~(-1)( Y =3.566X-0.006,r~2= 1.000); The recoveries were 95. 19% ~ 103. 70%. CONCLUSION The method is simple and accurate,and is suitable for determination of β-hydroxy-β-methylbutyrate in soy peptide protein complex powder.
