HPLC法测定清热消肿颗粒中秦皮乙素的含量

摘要(Abstract):

目的建立清热消肿颗粒中秦皮乙素的含量测定方法。方法色谱柱:Inert Sustain C_(18)柱(4.6 mm×150 mm,5μm),检测波长:350 nm,流速:1.0 m L·min~(-1),柱温:30℃,进样量:10μL。流动相:甲醇—0.1%磷酸水溶液(35∶65)。结果秦皮乙素浓度在1.016~16.256μg·mL~(-1)范围内,与峰面积的线性回归方程为:A=40.442C-2.301(r=0.999 9),线性关系良好。平均加样回收率为98.51%,RSD为1.3%。结论该方法准确、简便,可用于清热消肿颗粒中秦皮乙素的含量测定。
OBJECTIVE To establish a method for content determination of esculetin in QingRe Xiao Zhong granules. METHODS The samples were analyzed on a Inert Sustain C_(18)( 4.6 mm×150 mm,5 μm) column and the mobile phase consisted of methanol-0.1%phosphonic acid solution( 35 ∶65) at a flow rate of 1. 0 m L·min~(-1). The detection wavelength was set at 350 nm,the sample size was10 μL and the column temperature was 30 ℃. RESULTS The calibration curve was linear within the range of 1. 016~(-1)6. 256μg·m L~(-1)( r= 0.999 9) for esculetin.The average recovery was 99.51%,RSD was 1.3 %. CONCLUSION This method is accurate and rapid,can be used for determination of esculetin in QingRe Xiao Zhong granules.
