富硒青钱柳-黄精复方对高尿酸血症小鼠降尿酸作用研究

摘要(Abstract):

目的研究富硒青钱柳-黄精复方对高尿酸血症模型小鼠降尿酸作用的影响。方法采用氧嗪酸钾建立高尿酸血症小鼠模型,以生理盐水、阳性药别嘌呤醇和富硒青钱柳-黄精复方连续灌胃给药14 d。检测高尿酸血症小鼠血清尿酸(UA)、尿素氮(BUN)及肌酐(CR)水平,测定肝脏黄嘌呤氧化酶(XOD)、黄嘌呤脱氢酶(XDH)和腺苷脱氢酶(ADA)活力,HE染色观察肝脏、肾脏组织病理学变化。结果连续给药14 d后,适宜剂量的富硒青钱柳-黄精复方能显著降低高尿酸血症小鼠的UA、CR及BUN水平,抑制肝脏ADA、XOD及XDH酶活力,并能改善肝脏、肾脏组织的病理损伤程度。结论富硒青钱柳-黄精复方对高尿酸血症小鼠具有显著降尿酸功效,且肝肾不良反应较别嘌呤醇小。
OBJECTIVE To study the effect of the selenium-enriched Cyclocarya paliurus and Polygonatum sibiricum compounds on lowering uric acid in mice with hyperuricemia. METHODS The mice model of hyperuricemia were established with potassium oxazinate, and the hyperuricemia mice were administered with normal saline, positive drug allopurinol, and selenium-enriched Cyclocarya paliurus and Polygonatum sibiricum compounds for 14 days respectively. The serum uric acid(UA), urea nitrogen(BUN) and creatinine(CR) levels in hyperuricemia mice were detected, and liver xanthine oxidase(XOD), xanthine dehydrogenase(XDH) and adenosine dehydrogenase(ADA) vitality were determine. And the histopathological changes of liver and kidney were observed by HE staining. RESULTS After 14 days of continuous administration, the appropriate dose of the selenium-enriched Cyclocarya paliurus and Polygonatum sibiricum compounds reduced the levels of UA, CR and BUN in hyperuricemia mice significantly, inhibited the activity of liver ADA, XOD and XDH enzymes, and improved the degree of pathological damage to the liver and kidney tissue. CONCLUSION It shows that the selenium-enriched Cyclocarya paliurus and Polygonatum sibiricum compounds has significant effect on lowering uric acid in hyperuricemia mice, and the liver and kidney adverse reactions are less than allopurinol.
