HPLC-MS法分析珠芽蓼中的酚类成分

摘要(Abstract):

目的利用高效液相色谱-质谱联用技术(HPLC-MS)建立一种分析珠芽蓼中酚类成分的方法。方法采用50%甲醇超声提取制备样品;色谱柱为ZORBAX SB-Aq柱(150 mm×4.6 mm,5μm),流动相为0.1%甲酸水溶液和乙腈,梯度洗脱,流速0.8mL·min~(-1),检测波长275 nm检测有机酸类成分,360 nm检测黄酮类成分;离子阱质谱检测器用于化合物定性分析。采用"中药色谱指纹图谱相似度评价系统(2004A版)"对10批样品进行特征图谱相似度评价。结果从珠芽蓼样本中发现了20个色谱峰,质谱数据分析鉴定了13个成分,其中绿原酸类化合物为其主要酚类物质。10批样本的酚类成分特征图谱相似度在0.9以上。结论本实验建立的HPLC-MS分析方法,可以对珠芽蓼中的酚类成分进行综合分析,有助于提升珠芽蓼样品的质量控制标准。
OBJECTIVE To establish a method for analysis of phenol compounds in Polygonum viviparum by HPLC-MS.METHODS Samples were extracted by sonification with 50% methanol-water solution. Agilent ZORBAX SB-Aq column(150 mm×4.6 mm,5 μm) was used. Gradient elution buffer consisted of water(0.1% formic acid) and acetonitrile,flow rate was 0.8 mL·min~(-1).Organic acids were detected at 275 nm and flavonoids were detected at 360 nm. Ion trap mass spectrometry detector was utilized for qualitative analysis. Chromatographic data of samples were compared by Similarity Evaluation System(2004 A). RESULTS 20 chromagraphic peaks were found in sample and 13 phenol compounds were identified. The chlorogenic acid and its isomers were the major phenol components in Polygonum viviparum. The similarity coefficients of 10 samples were above 0. 9. CONCLUSION The developed HPLC-MS method is a comprehensive analytical method,which is helpful to the quality evaluation of Polygonum viviparum.
