黄芪多糖对糖尿病足溃疡成纤维细胞AGEs及RAGEmRNA表达的影响

摘要(Abstract):

目的研究黄芪多糖对糖尿病足溃疡成纤维细胞(Fbs)AGEs浓度及RAGE mRNA表达的影响。方法 30例2型糖尿病足溃疡患者随机分为黄芪多糖外敷治疗(A)组和外用生理盐水外敷治疗(B)组,各15例,组织块法培养Fbs,荧光分析法与逆转录一多聚酶链反应(reverse traBcription polymerase chain reaction,RT-PCR)技术检测成纤维细胞AGEs浓度及RAGEmRNA的表达,观察2组患者的肉芽组织出现时间(GT)和溃疡愈合时间(HT)。结果从第2周开始,A组的AGEs浓度及RAGEmRNA的表达显著低于B组(P<0.05),而且A组的GT和HT均显著低于B组(P<0.01)。结论黄芪多糖可以有效降低糖尿病足溃疡Fbs的AGEs浓度及RAGEmRNA的表达,显著缩短糖尿病足溃疡患者的GT和HT。
Objective To investigate the effects of astragalus polysaccharides on the concentration of AGEs and the expression of RAGEmRNA in human dermal fibroblasts derived from diabetic foot ulcers.Methods A total of 30 patients with type 2 diabetic foot ulcers were randomly divided into astragalus polysaccharides external application group(A) and physiological saline external application group(B),with 15 cases in each group.The tissue method was used to culture fibroblasts.The concentration of AGEs and the expression of RAGEmRNA were tested by fluorescence analysis and RTPCR.The time of granulation tissue appearance(GT) and ulcers healed(HT) of all the cases in two groups were also investigated.Results Starting from the second week,the concentration of AGEs and the expression of RAGEmRNA in group A were significantly lower than that in group B.GT and HT in group A were significantly lower than that in group B.Conclusion Astragalus polysaccharides can effectively reduce the concentrationof AGEs and the expression of RAGEmRNA in human dermal fibroblasts derived from diabetic foot ulcers and can significantly shorten the time of GT and HT of diabetic foot ulcers.
