北枳椇子与枳椇子UPLC指纹图谱的建立及特征成分分析Establishment of UPLC Fingerprints of Hovenia dulcis Thunb and Hovenia acerla Thunb and Analysis of Its Characteristic Components

摘要(Abstract):
目的建立北枳椇子和枳椇子超高效液相色谱(UPLC)指纹图谱,并用UPLC-Q-TOF-MS对特征成分定性分析。方法采用ACQUITY UPLC HSS T3色谱柱(100 mm×2.1 mm,1.8μm),以乙腈-0.2%甲酸水溶液梯度洗脱,流速0.15 m L/min,检测波长为340 nm;柱温25℃;飞行时间质谱采用负离子模式扫描。结果分别构建了北枳椇子和枳椇子药材UPLC指纹图谱共有模式,11个共有峰鉴定出12种黄酮类成分;其中北枳椇子的相似度均大于0.89,枳椇子的相似度除1批样品外,其余均大于0.81。结论方法准确可靠,重复性好,为药材质量控制提供了参考。
OBJECTIVE To establish the UPLC fingerprints of Hovenia dulcis Thunb and Hovenia acerla Thunb,and to identify the characteristic chemical components in the fingerprint by UPLC-Q-TOF-MS. METHODS The chromatographic fingerprint was obtained with ACQUITY UPLC HSS T3 column( 100 mm × 2. 1 mm,1. 8 μm) and gradient eluted with acetonitrile and 0. 2% formic acid; The flow rate was 0.15 m L/min,and the column temperature was maintained at 25 ℃. The detection wavelength was set at 340 nm. The mass spectrometer was operated in negative mode. RESULTS The UPLC fingerprints of Hovenia dulcis Thunb and Hovenia acerla Thunb were established to identify 12 compositions for 11 common peaks. The similarity of fingerprints of Hovenia dulcis Thunb was more than 0. 89 and the similarity of fingerprints of Hovenia acerla Thunb was more than 0. 81 except 1 batch of sample.CONCLUSION The method is accurate and reproducible,and it can provide a reference for the quality control of medicines.
