基于UPLC-Q-TOF-MS分析广金钱草水提物灌胃大鼠后肾组织中的化学成分及其代谢产物

摘要(Abstract):

目的对广金钱草在大鼠肾组织中分布的化学成分进行分析。方法采用UPLC-Q-TOF-MS对灌胃广金钱草提取物后的大鼠肾组织中的化学成分进行分析,色谱条件:流动相甲醇0.1%甲酸-水溶液梯度洗脱,流速0.4 mL·min~(-1),进样量1μL;质谱条件为电喷雾离子源,负离子模式扫描,扫描范围m/z 50~1 000。根据保留时间、精确相对分子质量、一级和二级质谱数据等鉴定和表征广金钱草在大鼠肾组织中的原型成分及代谢产物。结果广金钱草给药后,在肾组织中共表征出5个原型成分,代谢产物20个,其主要代谢途径为甲基化、葡萄糖醛酸化等。结论建立的方法可快速鉴定广金钱草的入肾原型成分及其代谢产物,有助于阐明广金钱草防治肾结石的潜在药效物质基础,为更好地了解广金钱草的体内吸收代谢机制提供参考。
OBJECTIVE To analyze the chemical components in rat renal tissues after oral administration of extracts of Desmodium styracifoLium. METHODS UPLC-Q-TOF-MS was used to analyze the renal tissues of normal rats after oral administration of the extracts of Desmodium styracifoLium.The chromatographic conditions were as follows: mobile phase methanol 0.1% formic acid aqueous solution gradient elution.The flow rate was 0.4 mL·min~(-1) and the injection volume was 1 μL.The mass spectrometry conditions were electrospray ion source, and negative ion mode scanning with scanning range m/z were 50-1 000.The prototype components and metabolites of Desmodium styracifoLium in rats were identified based on retention time, exact relative molecular mass, primary and secondary mass spectrometry data. RESULTS After the administration of the Desmodium styracifoLium,a total of 5 prototype components and 20 metabolites were identified in the kidney tissues.The main metabolic pathways were methylation, glucuronidation. CONCLUSION The method established in this experiment can be used to rapidly identify the protypes and metabolites in rat renal tissues, and provide a reference for better understanding of the mechanism of absorption and metabolism.
