微透析联用高效液相色谱法研究吴茱萸碱和吴茱萸次碱经皮吸收方法的建立
Establishment of EVO and RUT Transdermic Absorption Research Method by Microdialysis Combined with HPLC

摘要(Abstract):

目的建立吴茱萸碱(EVO)和吴茱萸次碱(RUT)的大鼠在体皮肤微透析方法。方法采用高效液相色谱法建立大鼠微透析样品中EVO和RUT的分析方法,考察药物浓度及微透析灌流液的温度、流速对体外回收率和体内回收率的影响。结果EVO、RUT在检测要求范围内线性关系良好,色谱的专属性、回收率、精密度等测定结果均符合生物样品测定要求。EVO、RUT的体外回收率与体外传递率无显著性差异,均与灌流温度成正比,灌流速度成反比,与药物浓度无关;最佳的在体皮肤微透析实验条件为灌流温度32℃,流速0.5μL/min。EVO、RUT经皮吸收药物浓度可采用体内回收率RL进行校正,EVO的体内回收率RL值分别为44.0%,RUT的体内回收率RL为30.50%。结论本方法能够在体、实时监测大鼠皮下EVO和RUT的动态浓度,为局部药动学研究提供新的方法学参考。
OBJECTIVE To establish a skin microdialysis method in vivo of EVO and RUT. METHODS An HPLC method was established for the determination of EVO and RUT in microdialysis samples,and the influence factors such as temperature,flow rate,and drug concentration on the probe recovery were investigated to obtain accurate data. RESULTS The results showed that the method of HPLC for the determination of EVO and RUT in microdialysis samples and plasma was both sensitive and reliable. Overall,the linearity correlation was good and the assay exhibited good precision and accuracy. The in vitro experiment results showed that in vitro recovery of EVO and RUT from the microdialysis probe determined by gain and by loss was comparable; in vitro recovery increased as the flow temperature increased; in vitro recovery decreased as the flow rate increased; EVO and RUT recovery was independent of concentration over the concentration range investigated. The optimal flow temperature was 32 ℃,the optimal flow rate was 0.5 μL / min.The in vivo recovery RL value of EVO was 44.0%; the in vivo recovery RL value of RUT was 30.50%. CONCLUSION The method can be used to determine in vivo and real time concentration of EVO and RUT in the skin dynamically. It is suitable for the pharmacokinetics study of EVO and RUT at the same time.
