基于核壳色谱技术快速测定珠芽蓼中3种有机酸的含量

摘要(Abstract):

目的建立珠芽蓼中新绿原酸、绿原酸和隐绿原酸快速含量测定方法。方法采用超声提取方法制备样品;使用核壳色谱柱-液相色谱法进行分析,色谱柱为PoroShell 120 EC-C_(18)(50 mm×4.6 mm, 2.7μm),流动相为0.1%乙酸水(A)和甲醇(B)梯度洗脱,柱温为35℃,检测波长为330 nm,流速为1.0 mL·min~(-1)。结果样品在5 min内完成了提取,在7 min内完成了色谱分离,3种有机酸成分线性关系良好(r>0.999 8);加样回收率99%~106%(RSD<1%)。15批样品3种有机酸的总含量在2.89~10.32 mg·g~(-1)之间,不同来源样品有机酸含量相差较大。结论本文首次建立了珠芽蓼中3种主要有机酸的含量测定方法,该方法快速、准确、可靠,为珠芽蓼的质量评价技术提升提供了参考。
OBJECTIVE To establish a quantitative method for rapid assay the contents of neochlorogenic acid, chlorogenic acid and cryptochlorogenic acid in Polygonum viviparum by HPLC. METHODS The sample extraction was prepared by ultrasonic method.Chromatographic separation was operated on a PoroShell 120 EC-C_(18) column(50 mm×4.6 mm, 2.7 μm) and gradient elution was performed by 0.1% acetic aqueous solution and methanol at a flow rate of 1.0 mL·min~(-1).The column temperature was maintained at 35 ℃.The detection wavelength was set at 330 nm. RESULTS The samples extraction were finished in 5 min and the sample separation was completed in 7 min.The calibration curve of 3 analytes showed good linearity(r>0.999 8) within tested range.The recoveries of 3 analytes were between 99% and 106%(RSD<1%).The total content of 3 organic acids in 15 batches of samples were between 2.89-10.32 mg·g~(-1).The contents of three organic acids in samples from different areas varied greatly. CONCLUSION In this study, a new HPLC method for determination of three main organic acids in Polygonum viviparum is established for the first time and it is a rapid, reliable and accurate method, which is help to improve the quality control of Polygonum viviparum.
