普伐他汀对大鼠脑血管平滑肌细胞增殖的影响

摘要(Abstract):

目的探讨普伐他汀(Pravastatin)对内皮素-1(ET-1)诱导的大鼠脑血管平滑肌细胞(basilar vascular smooth muscle cells,BVSMCs)增殖的比较。方法原代培养大鼠脑基底动脉平滑肌细胞,采用CCK-8(cell counting kit-8)、Brd U(5-bromo-2'-deoxyuridine)掺入实验检测普伐他汀和辛伐他汀和对ET-1诱导平滑肌细胞增殖的作用。结果普伐他汀和辛伐他汀均可以浓度依赖性地抑制ET-1诱导的BVSMCs增殖,相对吸光度百分比分别为:102.4%±8.9%,119.3%±9.8%,和对照组(132.5%±4.5%)相比差异均有显著性意义(P<0.01),普伐他汀组和辛伐他汀组比较二者差异也有显著性意义(P<0.01)。结论普伐他汀和辛伐他汀均可以抑制大鼠脑血管平滑肌细胞增殖,辛伐他汀的作用效果强于普伐他汀。
OBJECTIVE To investigate the inhibition effects of the pravastatin and simvastatin on the proliferation of rat vascular smooth muscle cells induced by ET-1. METHODS CCK-8( cell counting Kit-8) and Brd U( 5-bromo-2'-deoxyuridine) were administered to detect simvastatin and pravastatin on primary cultured basilar artery smooth muscle cells( BASMCs) proliferation in rats induced by ET-1 function. RESULTS Pravastatin and Simvastatin may be concentration-dependent inhibition of ET-1 induced BASMCs proliferation,the relative absorbance percentage were:( 102. 4% ± 8. 9%) vs( 119. 3% ± 9. 8%),compared with the control group( 132.5% ±4.5%),differences were significant( P < 0. 01) and pravastatin group. The difference of comparison between the 2groups was significant( P<0.01). CONCLUSION Pravastatin and simvastatin inhibits proliferation of basilar artery smooth muscle cells in rats,simvastatin effect stronger than pravastatin.
