过岗龙多酚正交试验优化提取的工艺研究Optimization of Extracting Polyphenols from Enteda phaseoloides by Orthogonal Test

摘要(Abstract):
目的研究过岗龙多酚的最佳提取工艺。方法以没食子酸为对照品,采用福林酚法测定多酚含量,采用单因素试验考察药物粉碎度、乙醇体积分数、料液比、提取时间及提取次数对过岗龙多酚提取效果的影响。在此基础上,采用L_9(3~4)正交试验优选过岗龙多酚提取工艺。结果最佳提取工艺:2 cm粒径过岗龙、6倍量水、60%乙醇回流提取2次,每次90 min。在此条件下,过岗龙提取得到的多酚含量为34.59%。结论该含量测定方法简便、准确、重复性好;提取工艺经济、合理。
To study the best extraction process of polyphenols from Entada phaseoloides( L.) Merr. METHODS The content of polyphenols was determined by Folin-Ciocalteu colorimetry with gallic acid as reference. Variables including herbal medicine particle size,ethanol concentration,solid-liquid ratio,extraction time extraction cycles were investigated by the single factor experiments,and extraction technology was optimized by orthogonal test. RESULTS The optimal conditions were: 2cm particle size of Entada phaseoloides( L.) Merr; 6 times volume; 60% ethanol reflux extraction 2 times,each time 90 minute. Under these optimal extraction conditions,the content of polyphenol reached 34.59%. CONCLUSION The established method of content determination is simple,feasible,highly accurate and economical.
