川芎嗪与阿米洛利联用对肝星状细胞增殖及分泌细胞外基质的影响

摘要(Abstract):

目的探讨钠氢交换抑制剂阿米洛利(Amiloride)与抗氧化剂川芎嗪联用对大鼠肝星状细胞(hepatic stellate cells,HSC)增殖及分泌细胞外基质的影响。方法用不同浓度的阿米洛利和川芎嗪处理大鼠肝星状细胞;以MTT比色法测定细胞的增殖;放射免疫法测定透明质酸和层黏连蛋白;酶联免疫吸附法(ELISA)测定Ⅰ型胶原水平。结果阿米洛利和川芎嗪均可剂量依赖性地抑制HSC增殖,不同程度抑制Ⅰ型胶原、透明质酸和层黏连蛋白分泌,联用组优于单用组。结论作用于HSC不同靶点的抗纤维化药物联用优于单用。
Objective To investigate the effects of antioxidant Chuanxiongzine and Na+/H+ exchanger inhibitor Amiloride on proliferation and extracellular matrix secretion of rat hepatic stellate cells in vitro. Methods HSC-T6 were incubated with different concentrations of Chuanxiongzine and Amiloride for 24 h. MTT colorimetry was used for assaying proliferation of HSC. Cytotoxicity was measured by LDH colorimetry. Type Ⅰcollage was determined by ELISA. Hyaluronic Acid and Laminin were measured by Radioimmunoassay. Results 1~20 mol/L Chuanxiongzine and Amiloride added in culture medium had no cytotoxicity to HSC, but could significantly inhibite HSC proliferation, and also inhibite the secretion of Type Ⅰ collage. Combining Chuanxiongzine with amiloride had better effects on proliferation and extracellular matrix secretion of rat hepatic stellate cells than single Chuanxiongzine or amiloride alone. Conclusion The effects of combination of antifibrosis drugs on different targets of HSC are superior to those of each one.
