急性酒精暴露对小鼠脑组织氨基酸以及神经递质含量的影响

摘要(Abstract):

目的通过研究急性酒精暴露对小鼠脑组织氨基酸以及神经递质水平的影响,探究酒精中毒的生理学基础。方法构建急性酒精暴露小鼠模型,通过亲水色谱串联质谱技术(HILIC/QQQ MS),同时对小鼠脑组织15种氨基酸以及神经递质的含量进行测定,对比实验组以及对照组代谢物水平的差异。结果急性酒精暴露小鼠脑组织色氨酸水平显著升高,丙氨酸、丝氨酸、甘氨酸以及谷氨酸水平均显著降低,差异具有统计学意义(校正后P<0.05)。结论本研究从脑组织水平证明了急性酒精暴露显著影响色氨酸、丙氨酸、丝氨酸、甘氨酸以及谷氨酸水平,除了丙氨酸外其余4种氨基酸均能影响谷氨酸能神经传递系统的活动,推测急性酒精暴露能够通过影响多个通路抑制谷氨酸能神经传递系统的神经传递。
OBJECTIVE To investigate the effect of acute alcohol exposure on various amino acids and neurotransmitters in mouse brain tissue,and to explore the pathophysiology of alcoholism basis. METHODS The mouse model of acute alcohol exposure were built. The hydrophilic interaction chromatography-tandem mass spectrometry( HILIC-MS/MS) method were used to determined 15 amino acids and neurotransmitters to find out the different metabolites between test and control group. RESULTS The levels of tryptophan of brain tissue was significantly higher in the test group. The levels of alanine,serine,glycine and glutamate of brain tissue in test group were significantly lower than that in controls. and the difference was statistically significant( adjusted P values less than0.05). CONCLUSION The result suggests that acute alcohol exposure can significantly change the amino acids and neurotransmitters,such as the alanine,serine,glycine,glutamate and tryptophan. Except for the alanine,serine,glycine,glutamate and tryptophan can affect the activity of glutamatergic system. We assume that acute alcohol exposure can inhibit the activity of glutamatergic system through various path ways.
