正交设计法优选蒲桃中总黄酮的提取工艺Extraction Technology of Total Flavonoids in Syzygium Jambos( L.) Alston by Orthogonal Design

摘要(Abstract):
目的利用正交设计法优选蒲桃中总黄酮的提取工艺。方法以芦丁为对照,硝酸铝为显色剂,用紫外分光光度法在510 nm处测定总黄酮的含量。采用L9(34)正交实验设计,以蒲桃中总黄酮含量为考察指标,考察乙醇浓度、提取时间、料液比、提取温度4个因素对提取的影响。结果最佳提取工艺为60%乙醇,70倍量,80℃,回流提取2 h。结论该工艺经试验重复性好,对进一步的工艺开发有较好的参考价值。
OBJECTIVE To optimize extraction technology of total flavonoids in Syzygium jambos( L.) Alston.METHODS The ultraviolet spectrophotometry method was used to measure the total flavonoids content in ethanol extract of Syzygium jambos( L.) Alston with different polar solvent extraction,with aluminium nitrate as color reagent and the rutin as standard substance and the maximum absorption wave-length of 510 nm. By L9( 34) orthogonal experimental design,with Syzygium jambos( L.) Alston,as index of total flavonoids were investigated ethanol concentration,extraction time,ethanol dosage and extraction temperature four factors on the extraction. RESULTS The optimum extraction process was 60% ethanol,70 times the amount of reflux extraction,80 ℃,and 2 h.CONCLUSION The experimental process has good reproducibility,and can provide a better reference for the further development of the process.
