广东狼毒中脯氨酸的含量测定

摘要(Abstract):

目的分析广东狼毒中氨基酸成分,为其质量控制提供重要的研究参数。方法采用HPLC-UV测定广东狼毒中脯氨酸的含量,流动相A:0.05 mol/L乙酸钠水溶液(冰乙酸调节pH值为6.50±0.05),流动相B:甲醇∶乙腈∶水=20∶60∶20(V∶V∶V),梯度洗脱;柱温:35℃,流速:1 ml/min,检测波长:254 nm,进样量:20μl。结果脯氨酸进样量在0.136 4~0.818 4μg范围内与峰面积呈良好的线性关系,平均回收率为98%,RSD(n=6)为1.4%。结论本含量测定方法分离效果好、灵敏度高、重复好,可作为广东狼毒氨基酸成分的测定方法。
Objective To analyze constituents of amino acide in Alocasia macrorrhiza,so as to provide important study parameters for its quality control.Methods HPLC-UV was used to determine the content of proline in Alocasia macrorrhiza.The eluent A was 0.05 mol/L sodium acetate(adjusted to pH 6.50±0.05 with glacial acetic acid),the eluent B was consisted of Methanol-acetonitrile-water(20∶60∶20).The column temperature was 35 ℃,the flow rate was 1.0 ml/min,and the detection wavelength was 254 nm.The sample load was 20 μl.Results The method had a good liner relationship while the sample load was between 0.136 4 and 0.818 4 μg,the average recovery rate was 99%,RSD(n=6) was 1.4%.Conclusion The method has good effect of separation,high sensitivity,and good repeatability,and can be used to determinate constituents of amino acide in Alocasia macrorrhiza.
