复方贞术调脂方治疗老龄脂肪萎缩小鼠脂肪肝的药理研究

摘要(Abstract):

目的观察复方贞术调脂方(FTZ)对老龄脂肪萎缩小鼠脂肪肝的影响以及脂肪组织在其中的作用。方法采用16月龄脂肪萎缩转基因小鼠(a P2-SREBP~(-1)c)为研究模型,该模型糖脂代谢紊乱伴有明显的脂肪肝,连续给予FTZ治疗12周,检测血浆、肝脏的总胆固醇(TC)和甘油三酯(TG)水平;给药12周后进行口服糖耐量实验(OGTT),计算曲线下面积(AUC);HE染色、油红O染色观察肝组织病理形态;采用气相色谱-质谱联用(GC-MS)测定肝脏脂肪酸含量。结果与对照组比较,模型组表现出明显的糖脂代谢紊乱,糖耐量异常,血浆及肝脏组织中TC及TG含量显著升高(P<0.05),肝脏脂肪酸水平明显增加,伴有严重的脂肪肝;与模型组比较,FTZ能显著调节血浆TC及TG的含量(P<0.05),降低肝脏中脂肪酸水平,对肝脏脂肪样变性有一定的改善作用。结论 FTZ对老龄脂肪萎缩小鼠脂肪肝有一定改善作用,其中脂肪组织发挥了重要作用。
OBJECTIVE To observe the effect of Compound Zhenzhutiaozhi Recipe( FTZ) on fatty liver in aging lipoatrophy mice and the role of adipose tissue in it. METHODS 16 months old transgenic mice with lipoatrophy were used as model. The model was characterized by lipid metabolism disorder and fatty liver. After treatment witn FTZ for 12 weeks,the levels of total cholesterol( TC) and triglyceride( TG) in both plasma and liver were tested. The oral glucose tolerance test( OGTT) was performed and the area under the curve( AUC) was calculated. The HE and oil red staining of liver were observed and the fatty acids in liver were determined by Gas Chromatography-Mass Spectrometry( GC-MS). RESULTS The model group showed obvious disorder of glucose and lipid metabolism and impaired glucose tolerance. Compared with the control group,the content of TC and TG in both plasma and liver tissue of the model group were significantly higher than the control group( P<0.05),and the total fatty acids level in the liver was significantly increased,showing a serious fatty liver. Compared with the model group,FTZ could significantly regulate the content of TC and TG in plasma,and lower the content of total fatty acids in liver( P<0.05). There was a certain improvement in liver adipose degeneration. CONCLUSION FTZ has a certain effect on improving fatty liver in aging mice with lipoatrophy,in which the adipose tissue plays an important role.
