尿石汤配方颗粒对大鼠草酸钙结石肾脏的保护作用

摘要(Abstract):

目的探讨尿石汤配方颗粒对大鼠草酸钙结石肾脏的保护作用。方法建立草酸钙大鼠肾结石模型,检测尿石汤不同剂量给药后大鼠血清中肌酐(CRE)、尿素氮(BUN)、Ca2+、Mg2+的变化以及肾脏组织病理变化和肾脏骨桥蛋白(OPN)的表达水平变化。结果造模后大鼠血清CRE、BUN、Ca2+水平显著升高,Mg2+水平显著下降,给药尿石汤配方颗粒后,大鼠血清中CRE、BUN、Ca2+水平呈下降趋势,Mg2+水平呈上升趋势。病理镜检,发现肾结石可造成肾脏肾小管显著扩张,草酸钙晶体大量沉积,给药后大鼠肾脏肾小管扩张程度明显降低,草酸钙晶体分布减少。模型组大鼠肾组织OPN显著表达,给药后,大鼠肾组织OPN表达见不同程度降低。结论尿石汤配方颗粒对大鼠草酸钙结石肾脏具有良好的保护作用,其原因可能为游离状态的OPN抑制草酸钙晶体核心的形成、聚集和成长,减少草酸钙晶体在肾脏的沉积,达到保护肾脏的作用。
OBJECTIVE To explore the protective effect of Niaoshitang formula granules on the kidney of rats with calcium oxalate stones. METHODS Calcium oxalate kidney stones rat model was to establish. The changes of creatinine( CRE),urea nitrogen( BUN),Ca2+,Mg2+in rat serum,the pathological changes of kidney tissue and renal osteopontin( OPN) expression level changes were detect after the model rats treated by different dose of Niaoshitang formula granules. RESULTS The levels of CRE,BUN,and Ca2+were increased significantly in the serum of rats with calcium oxalate stones,and the level of Mg2+was decreased significantly. After administration of the formula granules,the levels of CRE,BUN,and Ca2+in the serum of rats were showed a downward trend,and the level of Mg2+was showed an upward trend. Pathological microscopy revealed that kidney stones could cause the renal tubules to expand significantly,and calcium oxalate crystals were deposited in large amounts. After administration,the renal tubules in rats were significantly dilated and the distribution of calcium oxalate crystals decreased. In the model group,OPN was significantly expressed in the kidney tissues of the rats. After administration of the formula granules,the expression of OPN in the kidney tissues of the rats decreased with varying degrees. CONCLUSION The Niaoshitang formula granules have good protective effect on kidneys of rats with calcium oxalate stones. The reason may be that free OPN inhibits the formation,aggregation and growth of calcium oxalate crystal cores,reduce the deposition of calcium oxalate crystals in the kidney,and protect the kidney.
