金莲花软胶囊对人冠状病毒OC43的抑制作用

摘要(Abstract):

目的研究金莲花软胶囊(JS)对人冠状病毒OC43(OC43)体外感染的影响。方法采用噻唑蓝(MTT)法检测JS对于HRT-18细胞的安全剂量,采用反转录的聚合酶链式反应(RT-PCR)法检测JS给药前后对OC43体外感染的抑制作用。结果 JS在1 000μg·mL(-1)及以下浓度对HRT-18细胞无明显抑制作用,JS在800μg·mL(-1)及以下浓度对HRT-18细胞无明显抑制作用,JS在800μg·mL(-1)及400μg·mL(-1)及400μg·mL(-1)浓度下能明显抑制OC43在HRT-18细胞内的复制,抑制病毒感染引起的白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)表达的上升。结论 JS可以明显抑制OC43的体外感染。
OBJECTIVE To investigate the effects of Jinlianhua soft capsule(JS) on the infection of human coronavirus OC43 in vitro. METHODS The MTT method was used to detect the safe dose of JS for HRT-18 cells. The reverse transcription-polymerase chain reaction(RT-PCR) method was used to detect the inhibitory effect of JS against OC43 infection. RESULTS JS showed no significant inhibitory effect on HRT-18 cells at the concentrations of 1 000 μg·mL(-1)浓度下能明显抑制OC43在HRT-18细胞内的复制,抑制病毒感染引起的白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子-α(TNF-α)表达的上升。结论 JS可以明显抑制OC43的体外感染。

JS could inhibit the replication of of OC43 in HRT-18 cells at 800 μg·mL(-1) and 400 μg·mL(-1) and 400 μg·mL(-1) significantly, and inhibit the increase in the expression of cytokines(IL-1β, IL-6, TNF-α) caused by virus infection. CONCLUSION JS can significantly inhibit the in vitro infection of OC43.
