地鳖和金边土鳖的色谱及分子生物学鉴别研究

摘要(Abstract):

目的通过色谱及分子生物学建立地鳖和金边土鳖的鉴别方法。方法运用薄层色谱及基于线粒体基因细胞色素C氧化酶亚基Ⅰ(COⅠ)的DNA条形码对地鳖和金边土鳖进行鉴别。结果金边土鳖样品薄层色谱均有3个红色斑点,而有一批地鳖样品色谱在相应位置也有2个红色斑点,薄层色谱方法难以区分两者;地鳖与金边土鳖的DNA条形码通过基因数据库进行结果判定,比对结果与各批样品对应物种相同,成功区分两者。结论本研究建立的DNA条形码方法准确、稳定、重现性良好,为鉴别地鳖和金边土鳖提供科学可靠的方法。
OBJECTIVE To establish method of identification between Eupolyphaga sinensis Walker and Opisthoplatia orientalis Burm. by chromatography and molecular biology. METHODS Eupolyphaga sinensis Walker and Opisthoplatia orientalis Burm. were identified by TLC and the DNA barcoding based on mitochondrial gene cytochrome C oxidase subunit Ⅰ( COⅠ). RESULTS TLC showed that three red spots from all of the Opisthoplatia orientalis Burm. samples and two from a branch of the Eupolyphaga sinensis Walker samples were observed in the corresponding area,which was difficult to distinguish between these two cockroaches. By searching the gene database with the sequences generated from the samples,DNA barcoding method identified the species of Eupolyphaga sinensis Walker and Opisthoplatia orientalis Burm. correctly. CONCLUSION The established DNA barcoding method is accurate,stable and reliable,which can be applied to the identification between Eupolyphaga sinensis Walker and Opisthoplatia orientalis Burm.
