仙鹤草药物血清对人肺癌细胞A549增殖凋亡的影响

摘要(Abstract):

目的观察仙鹤草药物血清对人非小细胞肺癌细胞株A549增殖、形态、凋亡的影响,验证仙鹤草的体外抗肿瘤作用。方法不同方法制备仙鹤草含药血清,分别作用于A549细胞,通过CCK8的方法考察细胞增殖情况,选出最佳方法的仙鹤草含药血清;倒置显微镜观察最佳方法的仙鹤草含药血清对A549细胞形态的影响,并用Annexin V-FITC/PI双染流式细胞术考察其诱导细胞凋亡的作用。结果两种仙鹤草含药血清对A549细胞均有增殖抑制作用,与正常对照组比较其中含药血清Ⅱ效果更为明显(P<0.01),且能够促进细胞的早期凋亡和坏死,降低其存活率。结论仙鹤草含药血清有一定的体外抗肿瘤细胞生长作用,该作用受制备方法、剂量等因素影响。
OBJECTIVE To observe the effect of serum containing Agrimonia pilosa Ledeb on A549 cell proliferation,morphology,and apoptosis in order to verify Agrimonia pilosa Ledeb anti-tumor effect in vitro. METHODS Different methods were adopted for preparing serum containing Agrimonia pilosa Ledeb,which then was administrated to A549 cells,and cell proliferation was investigated by CCK8 method; inverted microscope was used for cell morphology; Annexin V-FITC / PI double dye flow cytometry was used for apoptosis. RESULTS Serum containing Agrimonia pilosa Ledeb from two methods both can inhibit proliferation of A549 cells,and compared with normal group,Agrimonia pilosa LedebⅡ had the better effect( P<0.01),which also significantly promoted cell early apoptosis and necrosis,reduced its survival. CONCLUSION Serum containing Agrimonia pilosa Ledeb has anti-tumor effect on A549 cells in vitro,which is related to its preparation method,dosage,and other factors.
