小活络丸的抑制免疫抗氧化抗炎及镇痛作用
Pharmacological effects of Xiaohuoluo Pill on immunosuppression, antioxidation, anti-inflammation and acesodyne in mice

摘要(Abstract):

目的 研究小活络丸对小鼠再次免疫应答、特异性免疫、非特异性免疫和自由基损伤,以及对疼痛及多种急性渗出性炎症模型、琼脂肉芽组织增殖性炎症模型的药理作用。方法 以两次腹膜内注射鸡红细胞(CRBC)诱导免疫增强病理模型探讨再次免疫应答;以2,4-二硝基氟苯(DNFB)诱导由1型T辅助性细胞以及巨噬细胞和B细胞联合介导的迟发型超敏反应(DTH)模型探讨细胞免疫;以碳粒廓清试验探讨MPS功能;以红细胞免疫粘附作用探讨红细胞免疫功能;以CRBC诱导小鼠病理模型的特异性抗体IgM溶血素活性(HC_(50)值)、丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性评价特异性体液免疫功能和抗自由基损伤能力;以及二甲苯、巴豆油和角叉菜胶致急性渗出性炎症模型、琼脂肉芽组织增殖性炎症模型及醋酸性疼痛模型;验证小活络丸的药效学效应。结果 小活络丸能降低再次免疫应答中高值的IgG和循环免疫复合物(CIC)含量和提高低下的C_3水平(P<0.05~0.01);抑制DTH、MPS的吞噬功能和红细胞免疫粘附功能(P<0.01);抑制CRBC诱导小鼠特异性抗体IgM型溶血素活性(P<0.01),提高C_3水平(P<0.01),降低MDA含量(P<0.01),对SOD活性则未见显著改变(P>0.05);并显示抗增殖性炎症、镇痛等的药理作用(P<0.05~0.01);短疗程给药,则未见抑制急性渗出性炎症水肿反应(P>
Objective To investigate pharmacological effects of Xiaohuoluo pill on imniunoenliancing anainnestic response to cock red blood cell ( CRBC ) , specific immune function ( comprising cellular and humoral immunity ) and nonspecific immune function [comprising complement 3 (C3) , red blood cell (RBC) immunity, mononuclear phagocyte system (MPS) ] and antioxidation, ache, various acute exuding and subacute proliferous inflammatory models in mice. Methods To observe the effects of Xiaohuoluo pill on levels of IgG and C3, circulating immune complexes (CIC) in twice intraperitoneally injected CRBC induced-secondary response; cellular immunity for 2 ,4-dinitrofluorobenzene (DNFB) induced T helper I cell, macrophage and B lymphocyte mediated-delayed type hypersensitivity (DTH) ; RBC immune adhering ability; activity of anti-CRBC specific antibody IgM (expressed 50% haemolytie concentration, HC50 value) and level of C3 and malondialdehyde ( MDA) , antioxidizing activity of superoxide dismutase (SOD) in mice immunised with CRBC; phagocytic function of MPS for test cleaning out Indian ink; and various inflammatory models in mice that dimethylbenzene or croton oil induced-acute exuding inflammatory swelling ears; carrageenin induced-acute exuding inflammatory swelling paws and increased content of prostaglandin ( PG ) Es; agar induced-granulation tissue proliferous inflammation, intraperitoneally injected acetic acid induced-aching response were studied. Results The effects were shown that levels of IgG, CIC were lowered and C3 was heightened ( P < 0. 05 -0.01 ) in secondary response; cellular immunity was inhibited ( P < 0. 01 ) ; RBC immune adhering ability was counteracted ( P < 0. 01 ) ; levels of MDA , C, and HC50 value were very significantly decreased ( P < 0. 01 ) and the activity of SOD was not changed (P > 0. 05 ) ; ability in cleaning out Indian ink of MPS was suppressed (P < 0. 01 ) ; and in a short period of treatment the different varieties of acute inflammation were not prevented (P>0.05) , but the synthesized or secreted content of PGEs was reduced (P < 0. 01 ) ; proliferous inflammation was controlled ( P < 0. 05 ) ; threshold of pain was increased (P<0.05) ; but in comparison with Xiaohuoluo pill the above-mentioned pharmacological results were not so powerful as cyclophosphamide, prednisone or diclofenac each other ( P < 0. 05 ~ 0. 01 ). Conclusions Xiaohuoluo pill possesses immunosuppression, antioxidation, anti-inflammation, acesodyne. Therefore, the pill can be administered medicine to patients with autoimmune disease such as rheumatic arthritis and rheumatoid arthritis.
