摘要(Abstract)：

目的分析附子炮制品煎煮液中生物碱成分含量,评价炮制品对小鼠阳虚模型的影响,探讨生物碱成分含量与附子"回阳救逆"功效相关性。方法采用HPLC法测定附子4种炮制品(黑顺片、高温炮制品、高压炮制品、微波炮制品)煎煮液中6种生物碱含量;考察4种炮制品对阳虚小鼠环核苷酸系统的影响,测定小鼠血浆环磷酸腺苷(c-AMP)、环磷酸鸟苷(c-GMP)、内皮素(ET)及免疫器官胸腺、脾脏指数等指标,评价附子炮制品"回阳救逆"功效。结果 4种炮制品比较,高压附炮制品单酯型生物碱溶出量较高,双酯型生物碱含量较低;与模型组比较,附子4种炮制品高、中、低3个剂量下均能提高阳虚小鼠胸腺指数和脾脏指数(P<0.05),增加c-AMP含量(P<0.05),降低c-GMP含量(P<0.05),提高ET值(P<0.05),并随着剂量增加与指标值呈一定的相关性;与黑顺片组比较,4个炮制品高、中、低3个剂量组均没有显著的差异(P>0.05),但高压炮制品中阳虚小鼠胸腺指数和脾脏指数最大,血浆中c-AMP和c-AMP/c-GMP最高,c-GMP含量最低。结论附子高压炮制品"回阳救逆"功效较好,其作用可能与生物碱成分保留相关。
OBJECTIVE To analyze the content of alkaloids of decoction,and compare the impact of products on mice with yang deficiency,explore the correlation of the content of alkaloids and " Yang deficiency" effect. METHODS The method of HPLC was used to determine the content of 6 kinds alkaloids of the 4 groups of the drug product groups( Heishunpian group,high temperature product group,high pressure product group,microwave treatment product group). The effect of 4 kinds of processed products on nucleotide systems of mice with Yang deficiency was examined. The content of plasma cyclic adenosine monophosphate( c-AMP),cyclic guanosine monophos-phate( c-GMP),and the ratio of c-AMP and c-GMP,endothelinimmune( ET),organs thymus,spleen coefficient of mice were measured to evaluate the effect of the drugs. RESULTS Compared with the other 3 kinds of processed products,the dissolution of ester alkaloids of high-pressure processed products was higher,the dissolution of diester alkaloids was lower; compared with the model group,4 kinds of processed products of Aconiti Lateralis Radix Praeparata all could improve the thymus index and spleen index( P<0.05),increased c-AMP levels( P<0.05),reduced c-GMP levels( P<0.05),increase the content of ET value( P< 0. 05),and there was a certain correlation for indicators with increasing dose; compared with the Heishunpian group,4processed products group had no significant difference at the same dose,but the content of pressure processing group of thymus index and spleen index maximum,plasma c-AMP and c-AMP/c-GMP were the highest and c-GMP content was the lowest. CONCLUSION The high pressure processed products had better effect of "restoring Yang and saving from collapse",and its effect may be associated with the alkaloids reserved.
