夏枯草干膏粉12种常见金属元素含量测定
Determination of 12 Common Metallic Elements in Dry Powder of Prunella Vulgaris L.

摘要(Abstract):

目的为了解夏枯草对实验动物的影响,对制备的实验用夏枯草干膏粉中铬(Cr)、锰(Mn)、铁(Fe)、镍(Ni)、铜(Cu)、锌(Zn)、砷(As)、银(Ag)、镉(Cd)、锑(Sb)、钡(Ba)、铅(Pb)12种金属元素含量进行测定,为夏枯草药理毒理学评价提供科学依据。方法随机抽取夏枯草干膏粉3份,分别准确称量0.4 g。湿法消解样品,定容至10 m L。采用在线内标,建立电感耦合等离子体质谱(ICPMS)技术对样品常见金属元素的检测方法,并对样品金属元素进行检测。结果建立的ICPMS测定方法,常见金属元素线性相关系数均>0.999 0;方法准确度、精确度和稳定性均达到检测技术要求,茶叶与冻干榉木叶粉2种有证参考物质验证,方法准确度、精密度高。夏枯草干膏粉中检测的12种常见金属元素含量均在国家中药或食品的各种限量范围。结论选用的夏枯草及制备的夏枯草干膏粉中检测的12种常见金属元素不影响夏枯草药理毒理学评价。
OBJECTIVE To investigate the effect of Prunella Vulgaris L. on experimental animals,12 common metallic elements(Cr,Mn,Fe,Ni,Cu,Zn,As,Ag,Cd,Sb,Ba and Pa) in dry powder of Prunella Vulgaris L. were determined,so as to provide the scientific evidence for the pharmacological and toxicological evaluation. METHODS Three samples of dry powder of Prunella Vulgaris L. were randomly extracted and weighed into 0. 4 g individually. The weighed samples were wet digested with porous electric heat digestion device,and set the volume of each digestion product into 10 m L with 5% HNO3. Based on internal standard method,the method of analyzing samples was established. Common metal elements in digested sample solutions were analyzed by ICP-MS.RESULTS The linear correlative coefficients for the common elements were more than 0. 999 0. The accuracy,precision and stability of the method were detected,and they all conformed with the analysis standard. The method was validated with 2 certified reference materials,tea leaves and Lyophilised beech leave powder. The 3 samples were determined. Contents of 12 common metallic elements in dry powder of Prunella Vulgaris L. were within the national safety limits of traditional Chinese medicine or food. CONCLUSION 12 common metallic elements did not affect the evaluation on the pharmacological and toxicological results of the selected Prunella Vulgaris L. and its dry powder.
