川芎嗪注射液对大鼠急性脊髓损伤后脊髓组织中MDA和SOD水平的影响Effect of Ligustrazine Injection on MDA / SOD in Spinal Cord Tissue after Acute Spinal Cord Injury of Rats

摘要(Abstract):
目的观察川芎嗪注射液对急性脊髓损伤后脊髓组织中丙二醛(MDA)和超氧化物歧化酶(SOD)水平的影响。方法选用Wistar健康大鼠,参照Freeman-Wright改良WD法复制大鼠急性脊髓损伤模型,随机分为模型组(n=30)、中药组(n=30)和西药组(n=30),另设正常对照组(n=15);造模后30 min,中药组给予川芎嗪注射液200 mg·kg~(-1),西药组给予甲强龙注射液30 mg·kg~(-1),0.5 h后,各实验组于2 h、6 h、24 h、3 d、7 d时间点采用硫代巴比妥酸(TBA)比色法检测脊髓组织中的MDA、SOD的含量。结果与正常对照组比较,模型组在2 h、6 h、24 h时间点,MDA水平显著升高而SOD活性显著降低,差异有统计学意义(P<0.05);与模型组比较,中药组、西药组在各时间点的MDA水平显著降低而SOD活性显著提高,差异有统计学意义(P<0.05)。结论川芎嗪注射液能提高脊髓组织中SOD活性,并抑制MDA的生成,有明显的抗自由基损伤能力,对大鼠急性脊髓损伤早期具有保护作用。
To observe the effects of ligustrazine injection on the levels of malondialdehyde( MDA) and superoxide dismutas( SOD) in the spinal cord after acute spinal cord injury. METHODS Acute spinal cord injury model was established by freeman-Wright WD method in wistar rats. A total of 105 healthy rats were randomly divided into blank group( n = 15),model group( n= 30),control group( n= 30) and treatment group( n= 30). Ligustrazine injection was given for half an hour after successful model establishment. The contents of MDA and SOD in the spinal cord tissues were detected by TBA colorimetric method at 2 h,6 h,24 h,3 d and 7 d after modeling. RESULTS Compared with the model group,the levels of MDA in the control group and the treatment group decreased significantly at 2h,6h and 24 h( P<0.05). Compared with the model group,the activity of SOD in the control group and the treatment group was significantly increased( P<0. 05). CONCLUSION Ligustrazine injection could improve the activity of SOD and inhibit the production of MDA. It may have the ability to protect the spinal cord in the early stage of acute spinal cord injury.
