黄芪总苷对四氯化碳所致大鼠肝纤维化的防治作用Anti-Hepatic Fibrosis Effects of Astragalosides on Carbon Tetrachloride-Induced Hepatic Fibrosis in Rats

摘要(Abstract):
目的观察黄芪总苷(Astragalosides,AST)对四氯化碳(Carbon Tetrachloride,CCL4)所致大鼠肝纤维化的防治作用。方法 CCL4皮下注射制备大鼠肝纤维化模型,造模4周后,灌胃给药AST 20,40,80 mg·kg-1·d-1,连续用药9周后测定肝纤维化大鼠的肝、脾指数,血清GPT和GOT,肝组织中羟脯氨酸(HYP)含量,并对肝组织进行病理组织学观察。结果 AST高、中、低3种剂量均可明显降低CCL4升高的血清GPT和GOT水平,使CCL4升高的大鼠肝组织中HYP含量恢复,高剂量还可明显降低肝、脾指数,AST的中、高剂量组均可明显减轻肝组织纤维化和肝硬化的病理组织学变化。结论 AST有明显的抗CCL4性肝纤维化作用,其中AST高剂量组的疗效最为显著。
OBJECTIVE To observe the prophylactic effects of Astragalosides( AST) on carbon tetrachloride( CCL4)-induced hepatic fibrosis in rats. METHODS CCL4 was injected subcutaneously in rats to induced hepatic fibrosis rats model. AST( 20,40 and 80 mg·kg- 1·d- 1) was given intragastrically for 9 weeks after 4 weeks of the model induction. The liver and spleen indices,the serum GPT,GOT,and Hydroxyproline( HYP) content of liver homogenates were assayed. RESULTS The elevation of serum GPT,GOT,and HYP content of liver homogenates were decreased remarkably by AST treatment. High dose of AST also decreased liver and spleen indices. Hepatic necrosis and fibrosis induced by CCL4 were ameliorated remarkably by AST high and middle dose treatment.CONCLUSION AST has an anti-hepatic fibrosis effect on CCL4-induced liver fibrosis in rats,especially the high dosage.
