一测多评法同时测定牡蒿中3种酚酸类成分的含量

摘要(Abstract):

目的建立一测多评法同时测定牡蒿中3种酚酸类成分的含量。方法采用Agilent Zorbax C18(4.6 mm×150 mm,5μm)色谱柱,以乙腈-0.1%甲酸为流动相进行梯度洗脱,流速1.0 m L·min-1,柱温30℃,波长325 nm。以异绿原酸A为内参物,建立其与绿原酸和异绿原酸C的相对校正因子,实现一测多评。结果绿原酸、异绿原酸A和异绿原酸C分别在6.02~300.90μg·m L-1(R2=0.999 3)、12.36~617.52μg·m L-1(R2=0.999 6)和3.47~173.76μg·m L-1(R2=0.998 8)范围内呈良好的线性关系,平均加样回收率分别为96.7%、97.1%和98.9%,RSD分别为2.87%、3.18%和0.65%。一测多评法测定的结果与外标法的结果无显著性差异。结论该方法同时测定了牡蒿中3种酚酸类成分的含量,方法稳定且重现性好,为提高牡蒿的质量标准提供科学新方法。
OBJECTIVE To establish a scientific method for simultaneous determination of three phenolic acids in Artemisia Japonica by quantitative analysis of multi-components by single-marker( QAMS). METHODS Chromatographic column of Agilent ZORBAX C18( 150 mm×4.6 mm,5 μm) was used in a gradient elution with acetonitrile-0.1% formic acid solution.The flow rate was1.0 m L·min-1,the column temperature was 30 ℃ and the detection wavelength was 325 nm.The relative correlation factors of chlorogenic acid and isochlorogenic acid C were determined by using isochlorogenic acid as the internal reference substance. RESULTS Chlorogenic acid,isochlorogenic acid A and isochlorogenic acid C showed a good linear relationship in the range of 6.02-300.90 μg·m L-1( R2= 0.999 3),12.36-617.52 μg·m L-1( R2= 0. 999 6) and 3. 47-173. 16 μg·m L-1( R2= 0. 998 8),respectively. The average recoveries were 96. 7%,97.1% and 98.9%,with RSD of 2.87%,3.18% and 0.65%,respectively. There was no significantly difference between QAMS and the external standard method( ESM). CONCLUSION A stable and reproducible method was developed to simultaneously determine the content of three ingredients,which can provide a scientific basis for improving the quality standards of Artemisia Japonica.
