三七总皂苷抗心肌细胞凋亡作用与MIF的关系研究

摘要(Abstract):

目的探讨三七总皂苷(panax notoginseng saponin,PNS)抗缺血诱导的心肌细胞凋亡的作用机制。方法将心肌H9c2细胞株分为正常对照组(control),模型组(model)和PNS组。Annexin V/PI染色检测细胞凋亡率,蛋白质免疫印迹法(Western blot)检测巨噬细胞迁移抑制因子(macrophage migration inhibitory factor,MIF)蛋白的表达水平。结果 PNS抗细胞凋亡的作用能够被MIF拮抗剂ISO-1显著逆转(P=0.000); Western blot结果显示PNS能够显著促进体外培养的H9c2细胞MIF蛋白的表达。结论 PNS抗心肌细胞凋亡的作用与激活MIF信号通路相关。
OBJECTIVE To investigate the mechanisms of the protective effect of panax notoginseng saponin (PNS) on apoptosis in H9c2 cells induced by SGOD (serum,glucose and oxygen deprivation). METHODS H9c2 cells were divided into normal control group,model group,and PNS group. Apoptotic cells were detected by flow cytometer (FCM) after stained with Annexin V/PI. MIF (macrophage migration inhibitory factor) protein expression level was detected by western blot assay. RESULTS FCM analysis shew that ISO-1,a MIF antagonist,could reverse the anti-apoptotic effect of PNS. Western blot assay demonstrated that SGOD significantly up-regulated the proteins expression of MIF,while PNS could further up-regulate the expression of MIF protein. CONCLUSION These results indicate that the mechanism of PNS inhibiting rat cardiomyocytes apoptosis induced by SGOD is involved with MIF signaling pathway.
