榴莲壳提取物propacin通过诱导自噬抑制脂多糖刺激的小鼠巨噬细胞炎症反应
Propacin Extracted from Durian Inhibits the Lipopolysaccharide-induced Inflammatory Response in Murine Macrophages by Inducing Autophagy

摘要(Abstract):

目的本课题组曾报道榴莲壳提取物propacin能有效抑制脂多糖诱导的小鼠巨噬细胞炎症反应,为了进一步探究其潜在的抗炎机制,本研究对propacin的抗炎活性与自噬的关系进行了探索。方法实验采用qR T-PCR及ELISA法检测了炎症相关因子的表达与释放情况,通过转染GFP-LC3质粒到小鼠巨噬细胞RAW264.7中观察LC3亮斑的形成情况,还通过Western blot法检测了自噬相关蛋白LC3的表达。结果 GFP-LC3亮斑的形成及LC3-I向LC3-Ⅱ的转化表明,propacin显著的增加了LPS诱导的RAW264.7细胞的自噬。qR T-PCR及ELISA结果显示propacin显著的降低了炎症相关因子IL-1β、IL-6、TNF-α、COX-2及iN OS的表达及TNF-α和IL-6的释放,而自噬抑制剂削弱了这种抑制作用。结论实验结果证明propacin通过增强自噬抑制了脂多糖诱导的RAW264.7细胞炎症反应。
OBJECTIVE To further explore propacin potential anti-inflammatory mechanism,and its relationship between the anti-inflammatory activity of propacin and autophagy based on previously reporte that propacin, a durian extract, suppressed lipopolysaccharide( LPS)-induced inflammation in murine macrophages. METHODS The expression and release of inflammation related factors were assessed by qRT-PCR and ELISA respectively.The formation of LC3 puncta were detected by transfection of GFPLC3 plasmid into RAW264. 7 murine macrophages cells,and the expression of autophagy related protein LC3 was also detected by western blot. RESULTS The formation of GFP-LC3 puncta and the conversion of LC3-I to LC3-Ⅱ revealed that propacin significantly increased LPS induced autophagy in RAW264.7 cells.The qRT-PCR and ELISA results showed that propacin significantly reduced the expression of inflammation related factor IL-1β,IL-6,TNF-α,COX-2 and iNOS expression and the release of TNF-α and IL-6,whereas autophagy inhibitors attenuated the inhibitory effect. CONCLUSION Our results demonstrate that propacin can suppress LPS induced inflammatory responses in RAW264.7 cells by enhancing autophagy.
