摘要(Abstract):
目的研究CCR5Δ32及CCR5 siRNA修饰的PBMC细胞对H IV-1假病毒的拮抗作用。方法将CCR5Δ32和CCR5 siRNA的基因连接到腺病毒载体上,用磷酸钙法将其转入人源性PBMC,W esternb lot检测CCR5Δ32稳定表达情况以及CCR5 siRNA对CCR5的抑制情况。构建H IV-1假病毒,检测荧光素酶报告基因的活性,判断病毒的感染情况。结果 CCR5Δ32在PBMC细胞上获得稳定表达,CCR5 siRNA抑制PBMC细胞中CCR5的表达;修饰后的PBMC细胞并不能被H IV-1假病毒感染。结论修饰后的PBMC细胞具有较好的拮抗H IV-1假病毒感染的作用。
Objective To detect the function of human peripheral blood-derived PBMCs modified by CCR5Δ32 and CCR5 siRNA to HIV-1 psedovirus.Methods The adenovirus-derived CCR5Δ32-CCR5siRNA vector was constructed.The genes were transfected into human peripheral blood-derived PBMC cells by using calcium phosphate.The HIV-1 psedovirus was constructed,and its infection was determined by testing the activity of luciferase.Results The stable expression of CCR5Δ32 in human peripheral blood-derived PBMC cells was obtained.CCR5 was inhibited by CCR5siRNA.Those modified PBMC cells could not be infected by HIV-1 pseudovirus.Conclusion Compared to normal PBMCs,the modified cells show good characteristic in resisting HIV-1 psedovirus infection.
