摘要(Abstract)：

目的探讨他克莫司滴眼液对小鼠结膜干燥综合征的治疗作用。方法选取健康的100只SD大鼠随机分为5组,除空白组外,其他各组小鼠采用免疫学方法建立小鼠结膜干燥综合征模型。空白组和模型组分别滴用空白对照液,3组给药组分别滴用他克莫司滴眼液、环孢素A滴眼液、地塞米松滴眼液。试验第1、4、7天检测各组酚红棉线长度、泪膜破裂时间、24 h饮水量;2周后取小鼠双眼结膜作病理切片观察腺体破坏程度和淋巴细胞浸润程度。结果与空白组比较,模型组与各给药组的酚红棉线长度以及泪膜破裂时间明显缩短(P<0.01),与模型组比较,他克莫司滴眼液组,环孢素A滴眼液组、地塞米松滴眼组酚红棉线长度以及泪膜破裂时间明显延长(P<0.05或P<0.01);组织病理学结果显示:他克莫司滴眼液对穹窿部上皮细胞增生的抑制作用明显强于地塞米松和环孢素A;他克莫司滴眼液对杯状细胞减少的抑制作用亦明显强于地塞米松滴眼液。结论他克莫司滴眼液对结膜干燥综合征有明显的治疗作用,且作用强于环孢素A滴眼液。
OBJECTIVE To observe the clinical effect of tacrolimus eye drops on mouse keratoconjunctivitis. METHODS One hundred SPF BALB / c mice were divided into 5 groups randomly( normal control group,keratoconjunctivitis model control group,tacrolimus eye drops group,cyclosporine A( Cs A) eye drops group,dexamethasone eye drops group). The keratoconjunctivitis mouse model was established with the method of immunological on all the groups except the control group. The phenol red thread length and the tear film break-up time and 24 h drinking volume were recorded at the 1,4 and 7 days after model establishment. After 2 weeks of consecutive drugging,the degree of damage and the degree of lymphocyte infiltration in mice with the pathological sections were observed.RESULTS The results showed that the phenol red thread length and the tear film break-up time of the 4 treatment groups were significantly shorter than the control group( P<0.01),and the phenol red thread and the tear film break-up time of tacrolimus eye drops group,Cs A group,dexamethasone drops group were significantly longer than the model group( P< 0.05 or P< 0.01). The fornix epithelial cell proliferation inhibition of tacrolimus drops group was significantly stronger than dexamethasone drops group and the Cs A drops group,and the inhibitory effect on goblet cells reduce of tacrolimus drops group was also significantly stronger than the dexamethasone drops group. CONCLUSION The tacrolimus eye drops have significant effect on keratoconjunctivitis,and the effect stronger than Cs A eye drops
