右美托咪定对脂多糖诱导HLF细胞凋亡的影响Effects of Dexmedetomidine on LPS-induced Apoptosis in HLF Cells

摘要(Abstract):
目的探讨不同浓度右美托咪定(Dex)对脂多糖(LPS)诱导人肺成纤维样细胞(HLF)凋亡的影响。方法实验分为5组:对照组、LPS 5μg·mL~(-1)组、LPS 10μg·mL~(-1)组、Dex 0.1 nmol·L~(-1)+LPS 10μg·mL~(-1)组、Dex 1 nmol·L~(-1)+LPS 10μg·mL~(-1)组;采用Hochest染色和caspase-3活性实验检测不同浓度LPS对HLF细胞凋亡的影响。结果 LPS 5μg·mL~(-1)、LPS 10μg·mL~(-1)均能够诱导HLF细胞产生凋亡小体,并增加caspase-3活性;Dex 0.1 nmol·L~(-1)、Dex 1 nmol·L~(-1)提前作用后,凋亡小体产生减少,且caspase-3活性显著降低(P<0.05)。结论右美托咪定在0.1~1 nmol·L~(-1)的浓度范围内,能够逆转LPS诱导的HLF细胞凋亡。
OBJECTIVE To investigate the effect of dexmedetomidine on LPS-induced apoptosis in HLF cells. METHODS The experiment was divided into 5 groups: control group,LPS 5 μg·mL~(-1) group,LPS 10 μg·mL~(-1) group,Dex 0.1 nmol·L~(-1)+LPS 10 μg·mL~(-1) group,and Dex 1 nmol·L~(-1)+LPS 10 μg·mL~(-1) group.Hochest staining assay and the caspase-3 activity were used to detect the effect of LPS on apoptosis in HLF cells. RESULTS Apoptotic bodies and an increase of caspase-3 activity were observed when treated with LPS 5 μg·mL~(-1)、LPS 10 μg·mL~(-1); pretreatment of 0.1-1 nmol·L~(-1) dexmedetomidine resulted in a decrease in numbers of apoptotic bodies and caspase-3 activity( P<0.05). CONCLUSION Dexmedetomidine reversed LPS-induced apoptosis at concentrations of 0.1-1 nmol·L~(-1) in HLF cells.
