土茯苓总黄酮的大孔吸附树脂纯化工艺Optimization of Purification Technology for Total Flavonoids in Smilax Glabra Roxb. by Macroporous Adsorption Resin

摘要(Abstract):
目的优选大孔树脂纯化土茯苓总黄酮的工艺条件。方法以土茯苓总黄酮的吸附率和解吸率为考察指标,考察11种大孔吸附树脂对土茯苓总黄酮的纯化性能,优选最佳性能的吸附树脂,确定纯化土茯苓总黄酮的最佳工艺参数。结果 HP20具有最佳的吸附分离性能,其纯化土茯苓总黄酮的工艺条件为:提取液上样质量浓度为0.72 g/L,上样流速为2.5 m L/min,洗脱剂为60%乙醇,洗脱量为6BV。纯化1次后,土茯苓总黄酮含量由粗提物的26.8%提高至79.7%。结论 HP20型大孔吸附树脂能更好地纯化土茯苓总黄酮,工艺条件为制备土茯苓总黄酮、研发土茯苓组成的复方中药提供物质基础。
OBJECTIVE To optimize the purification technology of total flavonoids from Smilax glabra Roxb. using macroporous adsorption resin.METHODS Adsorption and desorption rate of total flavonoids in Smilax glabra Roxb. was observed to examine for better conditions through optimization process by 11 types of macroporous adsorption resin.RESULTS HP20 had better adsorption and desorption rate. The purification process conditions were as follows: p H value of extract was 2.79,concentration of total flavonoids was0. 72 g / L,flow rate was 2.5 m L / min,elution solvent was 60% ethanol,and elution volume was 6 times column volume. After adsorption and desorption,the content of total flavonoids extraction from Smilax glabra Roxb. was increased from 26.8% to 79.7%. CONCLUSION HP20 macroporous adsorption resin has better abilities in purification flavonoid from Smilax glabra Roxb.,the optimized technology could provide flavonoid purification and materials for studying prescriptions composed of Smilax glabra Roxb.
