RP-HPLC测定人血浆中盐酸二甲双胍的浓度

摘要(Abstract):

目的以RP-HPLC测定人血浆中盐酸二甲双胍的浓度。方法色谱柱为D iamosil C18(5μm,4.6 mm×250 mm),流动相为0.01 mol.L-1KH2PO4-0.006 mol.L-1SDS-乙腈(34.25∶34.25∶31.5),流速为1.0 m l.m in-1,检测波长为233 nm,内标为阿替洛尔。结果盐酸二甲双胍的线性范围为0.05~4.00μg.m l-1,回归方程为Y=1.3397X+0.0222(r=0.9999);日内精密度为1.25%~6.14%,日间精密度为0.93%~6.43%。结论本方法操作简便,测定结果准确,重复性好,可用于盐酸二甲双胍人体药代动力学研究。
Objective To establish a RP-HPLC for the determination of Metformin Hydrochloride in human plasma.Method The Diamosil C_(18) column(5 μm,4.6 mm×250 mm) was used with mobile phase of 0.01 mol·L~(-1) KH_2PO_4-0.006 mol·L~(-1) SDS-acetonitrile(34.25∶34.25∶31.5).The flow-rate was 1.0 ml·min~(-1).The detection wavelength was 233 nm.Internal standard was Atenolol.Result The linear range of the calibration was(0.05~4.00) μg·ml~(-1)(Y=1.3397X+0.0222,r=0.9999).Within-day RSD was 1.25%~6.14%.Between-day RSD was 0.93%%~6.43%.Conclusion The method is simple,accurate and was used in the phamarmacokinetic study of Metformin Hydrochloride.
