微透析技术研究盐酸青藤碱在家兔体内的药动学特征Pharmacokinetic Studies of Sinomenine Hydrochloride by Microdialysis Technique In Ear Vein and Knee of Rabbits

摘要(Abstract):
目的探讨采用微透析技术研究盐酸青藤碱药动学的可行性及优越性。方法以家兔为实验对象,盐酸青藤碱静脉内给药,在耳缘静脉及膝关节部位植入探针,采用微透析技术取样,高效液相色谱法分析,3p87程序分析数据。结果青藤碱在家兔血液中的药-时曲线符合二室模型,在关节液中符合一室模型,在血液中,盐酸青藤碱达峰时间约为4 min,消除相半衰期约为55 min;在关节液中,盐酸青藤碱的达峰时间约为66 min,消除相半衰期约为196 min。结论采用微透析技术研究盐酸青藤碱药动学的方法可行,与传统的方法相比,具有明显的优势。
OBJECTIVE To explore the feasibility and advantages of microdialysis technique in the pharmacokinetic study of sinomenine hydrochloride.METHODS In this study,rabbits were served as the experimental subjects,and sinomenine hydrochloride was administered to the rabbits by intravenous injection.Probes were implanted in ear vein and knee of rabbits,and the blood was sampled by the microdialysis technique.Sinomenine hydrochloride was determined by high performance liquid chromatography(HPLC)and the associated pharmacokinetic parameters were analyzed by 3p87 program.RESULTS The drug-time curve of sinomenine in rabbit blood was consistent with the two-compartment model,and consistent with the one-compartment model in the joint fluid.The time to peak was 4 min in blood and 66 min in knee,the half-life of elimination phase was 55 min in blood and 196 min in knee.CONCLUSION It is feasible to study pharmacokininetic parameters of sinomenine hydrochloride by using microdialysis technique.In comparisom with the traditional technique,microdialysis technique is superior to the examination of drug blood concentration.
