甘草渣的质量标准研究

摘要(Abstract):

目的制定甘草渣药材质量标准,为甘草渣的合理开发利用提供科学依据。方法采用薄层鉴别法(TLC)对甘草渣进行定性鉴别,参照《中国药典》方法进行常规项目检查,以甘草苷为对照品,采用紫外分光光度法测定甘草渣中总黄酮的含量,采用高效液相色谱法测定甘草渣中甘草酸和甘草苷的含量。结果甘草渣药材薄层色谱中,在对照品相同位置显相同颜色斑点。甘草渣中水分平均含量为7.66%,灰分平均含量为7.94%,酸不溶性灰分平均含量为2.17%。紫外分光光度法表明甘草苷回归方程为:y=5.462x-0.008 3(r=0.999 7),回收率为100.74%(RSD=1.38%),5批甘草渣中总黄酮平均含量为1.81%。HPLC定量分析,甘草苷和甘草酸的回归方程为y甘草苷=14 314x+31.44(r=0.999 2);y甘草酸=3892.9x+21.473(r=1)。结果表明,甘草苷在0.222~1.11 mg/m L浓度范围内,甘草酸在0.36~2.16 mg/m L浓度范围内,峰面积与浓度呈良好的线性关系,平均回收率分别为100.74%和99.34%,RSD分别为1.38%和2.50%。不同批次甘草苷和甘草酸平均含量为0.019%和0.57%。结论所建立的分析方法简便、准确、可靠,具有很好的专属性和准确性,可用于甘草渣药材的质量控制。
OBJECTIVE To establish the quality standard system of residues of Glycyrrhiza for the further application and development.METHODS The contents of moisture,total ash and total insoluble ash were respectively measured according to the relative determination methods in appendix IX of Chinese Pharmacopoeia of 2010 edition.TLC method was adopted to qualitatively identify residues of Glycyrrhiza and quantitative determination included the determination of total flavoniods by UV and the content determination of liquiritin and glycyrrhizic acid by HPLC.RESULTS The contents of moisture,total ash and total insoluble ash were7.66%,7.94% and 2.17%,respectively.UV result showed the liquiritin linear correlation was y=5.462x-0.008 3(r = 0.999 7) and the average recovery was 100.74%(RSD=1.38%),the average content of total flavonoids in residues of Glycyrrhiza was 1.81%.HPLC result showed that the linear correlation of liquiritin and glycyrrhizic acid were y=14 314 x+31.44(r=0.999 2) and y=3 892.9x+21.473(r=1),respectively.The detection limits of liquiritin and glycyrrhizic acid were 0.222-1.11 mg/mL and 0.36-2.16 mg / m L,respectively.The average recovery of liquiritin and glycyrrhizic acid were 100.74%(RSD=1.38%) and 99.34%(RSD=2.50%),respectively.The average content of liquiritin and glycyrrhizic acid in residues of Glycyrrhiza were 0.019% and 0.57%,separately.CONCLUSION This method is convenient,accurate,reliable with good reproducibility,so it can provide scientific basis for establishing the quality standard of residues of Glycyrrhiza.
