白木香叶有效成分提取工艺优化研究

摘要(Abstract):

目的利用高效液相色谱(HPLC)法分析白木香叶有效成分提取工艺,确定最佳提取条件,为白木香叶试验研究及工业开发提供依据。方法通过单因素试验分别考察不同提取溶剂(甲醇与乙醇)、不同提取溶剂浓度(70%甲醇与甲醇)、不同提取方式(加热回流提取与超声提取)、不同提取时间(20,30,40 min)及料液比(0.25 g∶25 m L、0.25 g∶50 m L、0.5 g∶25 m L、0.5 g∶50 m L)对白木香叶成分提取效果的影响。结果 HPLC色谱图显示,甲醇提取的主成分峰比乙醇提取的主成分峰的响应值高,且出峰数量多; 70%甲醇与甲醇提取的供试品色谱图出峰数目基本无异;两种提取方式所得供试品色谱图出峰数目基本无异;时间对提取效果的影响不明显,但随着时间的延长,主成分峰的峰面积略有提高;不同提取料液比提取效果无明显差异。结论白木香叶的最佳提取条件确定为,70%甲醇作为提取溶剂、选择超声提取30 min及选择0.25 g∶25 m L的料液比。
OBJECTIVE To determine the best conditions for the extraction process of active constituents from Aquilaria Sinensis leaves by high performance liquid chromatography( HPLC),in order to provide the basis for experimental research and industrial development of it. METHODS Single factor experiment was carried out to investigate different extraction solvents( methanol and ethanol),different extraction solvent concentrations( 70% methanol and methanol),different extraction methods( heated reflux extraction and ultrasonic extraction),different extraction times( 20 min,30 min,40 min) and effect of solid-liquid ratio( 0.25 g ∶25 m L,0.25 g ∶50 m L,0. 5 g ∶ 25 m L,0. 5 g ∶ 50 m L) on the effects of extracting ingredients of Aquilaria Sinensis leaves. RESULTS The chromatograms of HPLC illustrated that the main component peak of methanol extraction had higher response value than that of ethanol,and the number of peaks was more. The number of peaks in the chromatogram of the test sample obtained by the extraction method was basically the same. The effect of time on the extraction effect was not obvious,but the peak area of the principal component peak increases slightly with time. There was no significant difference in the extraction efficiency of different extraction to liquid ratio.CONCLUSION The optimal extraction conditions of Aquilaria Sinensis leaves are determined as 70% methanol,ultrasonic extraction for 30 min and material to liquid ratio of 0.25 g ∶25 m L.
