珠三角地区人群血栓弹力图AA抑制率与PEAR1和GP1BA基因多态性相关性研究

摘要(Abstract):

目的为了探讨在阿司匹林个体化治疗中,血栓弹力图中AA抑制率与PEAR1(rs12041331)和GP1BA(rs6065)基因多态性是否具有相关性。方法在中山大学附属第五医院2018年1月至2020年12月间,招募了146例使用阿司匹林的患者。给予100 mg剂量3 d。然后根据血栓弹力图的AA抑制率,将患者分为低、适当和高药物反应组。同时,通过荧光原位杂交脱氧核糖核酸(DNA)测序系统,检测PEAR1和GP1BA的单核苷酸多态性。最后分析三组间基因多态性的统计学差异。结果 (1)阿司匹林药物反应性在不同性别没有差异。(2)PEAR1的GG、GA和AA频率分别为0.301、0.493和0.206,G和A等位基因频率分别为0.548和0.452;(3) GP1BA的CC、CT和TT频率分别为0.897、0.103和0,C和T等位基因频率分别为0.949和0.051;(4)PEAR1和GP1BA的单核苷酸多态性在高、适当和低组间差异无统计学意义。结论在中国人群的阿司匹林个体化治疗中,TEG的AA抑制率与PEAR1和GP1BA的基因多态性无关。
OBJECTIVE To explore whether the rate of AA inhibition of thrombelastogram is related with genetic polymorphisms of PEAR1(rs12041331) and GP1 BA(rs6065) in the individualized therapy of aspirin. METHODS 146 patients who used aspirin were recruited from January 2018 to December 2020 in the Fifth Affiliated Hospital of Sun Yat-sen University.100 mg dosage was used for 3 days.Then the patients were divided into low, proper and high drug response groups according to the AA inhibition of thrombelastogram.Meanwhile, the single nucleotide polymorphisms(SNPs) of PEAR1 and GP1 BA were determined by fluorescence in situ hybridization DNA sequencing systems.Finally the statistical differences of gene polymorphisms among three groups were analyzed. RESULTS(1) The response of aspirin did not differ between genders.(2) The GG,GA and AA frequencies of PEAR1 were 0.301,0.493 and 0.206,respectively.The G and A allele frequencies were 0.548 and 0.452.(3) The CC,CT and TT frequencies of GP1 BA were 0.897,0.103 and 0.The C and T allele frequencies were 0.949 and 0.051.(4) There was no statistical difference(P>0.999) among the high, proper and low groups in the single nucleotide polymorphism(SNP)of PEAR1 and GP1 BA. CONCLUSION The rate of AA inhibition of thrombelastogram is not associated with the gene polymorphisms of PEAR1 and GP1 BA in the individualized therapy of aspirin in Chinese populations.
