他克莫司滴眼液对碱烧伤兔干眼症疗效的实验研究
Study on Alkali Burn Dry Eye Treated with Tacrolimus Eye Drops in Rabbits

摘要(Abstract):

目的初步探讨他克莫司滴眼液对兔干眼影响。方法新西兰白兔18只(36只眼),制作碱烧伤干眼模型,左眼为空白对照组,右眼为造模眼。造模后随机分为3组,模型对照组、他克莫司滴眼液组和阳性对照药重组牛碱性成纤维细胞生长因子滴眼液组,每组6只眼。术后第1天起,各组给予相应的药物进行治疗,连续10 d。造模后第1,5,10天,对各组进行SchirmerⅠ试验、But试验、结膜荧光素染色和虎红染色检查。给药10 d后麻醉处死实验动物,取眼睑结膜及穹窿部组织进行病理学检查。结果给药第10天,与模型组相比,他克莫司滴眼液组酚红棉线湿润长度增加、荧光素和虎红染色评分明显降低,有统计学差异(P<0.05)。组织病理学也表明他克莫司对结膜损伤部位、结膜穹窿部位的充血和水肿也具有明显的改善作用。结论他克莫司滴眼液能改善兔干眼局部体征,从而为干眼的药物治疗提供了新的选择。
OBJECTIVE To investigate the effects of tacrolimus eye drops in dry eye rabbits. METHODS Eighteen rabbits( 36eyes) were used in this study. All left eyes were blank control group. Dry eye model was established by treating the conjunctiva with alkali. The rabbits were then randomly divided into 3 groups: model control group,tacrolimus eye drops group,and positive control b FGF eye drops group,with 6 eyes in each group. Since the 1st day after operation,the treatment group received respective treatments for 10 d. Schirmer I test,BUT test,corneal fluorescein staining and rose bengal staining were performed in 1,5,10 d after operation. At day 10 after treatment,the rabbits were sacrificed to observe the pathomorphism in the tarsal conjunctiva. RESULTS Compared with model control group,moistening length of filter paper increased significantly,the levels of corneal fluorescein and rose bengal staining decreased significantly in tacrolimus eye drops group at 10 th day( P<0.05). Histopathology suggested that the injury of conjunctiva and fornix was prevented by tacrolimus eye drops. CONCLUSION Tacrolimus eye drops can alleviate the symptoms of dry eye. As such,tacrolimus eye drops is a new way for the prevention and treatment of dry eye.
