珠芽蓼果实高效液相色谱指纹图谱研究

摘要(Abstract):

目的采用高效液相色谱法(HPLC)研究珠芽蓼果实的指纹图谱,为珠芽蓼果实质量评价提供参考。方法采用超声提取方法制备供试品溶液;利用HPLC法对珠芽蓼果实样品进行质量分析,色谱柱为Agilent ZORBAX SB-Aq(4.6 mm×250 mm, 5μm),流动相为乙腈(A)-0.2%磷酸水溶液(B),梯度洗脱,流速0.8 mL·min~(-1),柱温25℃,变波长检测:0~37 min检测波长275 nm, 37~80 min检测波长330 nm,进样量10μL;将获得的样品色谱图导入中药色谱指纹图谱相似度评价系统(2012版)构建指纹图谱并进行相似度评价;运用SPSS 16.0软件对15批样品进行聚类分析。结果从样品中发现了12个共有色谱峰,指认了其中的5个,分别为没食子酸、新绿原酸、儿茶素、绿原酸和芦丁;15批样品与参照图谱R的相似度在0.667~0.991之间;聚类分析可将15批样品分为3类。结论本研究所建立的分析方法稳定可靠,为珠芽蓼果实质量评价的提升提供参考。
OBJECTIVE Development of a high performance liquid chromatography(HPLC) fingerprint method for quality evaluation of Polygonum viviparum fruit. METHODS The sample solution was prepared by ultrasonic extraction.HPLC analysis of Polygonum viviparum fruit was operated on Agilent ZORBAX SB-Aq column(4.6 mm×250 mm, 5 μm), the mobile phase was acetonitrile(A)-0.2% phosphoric acid solution(B), gradient elution.The column temperature was set at 25 ℃ and flow rate was 0.8 mL·min~(-1).The components were detected by variable wavelength: 0-37 min detected at 275 nm, 37-80 min detected at 330 nm, the sample injection amount was 10 μL.The obtained chromatogram was imported into the similarity evaluation system(2012 edition) of traditional Chinese medicine chromatographic fingerprint to carry out the fingerprint similarity evaluation. SPSS 16.0 software was used for cluster analysis of samples. RESULTS In fingerprint analysis, 12 common chromatographic peaks were found in Polygonum viviparum fruit sample, and five of them were identified as gallic acid, neochlorogenic acid, catechin, chlorogenic acid and rutin; the similarity values of 15 batches of samples were between 0.667 and 0.991.In cluster analysis, 15 batches of samples can be divided into 3 categories. CONCLUSION The established method was stable and reliable, which was helpful to improve the quality evaluation of Polygonum viviparum fruit.
